We use live transgenic zebrafish expressing green fluorescent protein under the control of gene-specific promoters to develop animal models of human disease to examine pharmacological activities and questions of gene function. Primary focus is on nicotinamide adenine dinucleotide (NAD) precursors (nicotinic acid/niacin, nicotinamide/niacinamide, nicotinamide riboside, or tryptophan), NAD salvage pathway enzymes, and the associated non-redox NAD-dependent families of proteins: Sirtuins (SIRT1-7) and poly(ADP)ribose polymerases (PARP1 to estimates of 18). These are most often related to cytotrophic activities. Our research is most clinically relevant to mental health, obesity, and cardiovascular disease. Caloric-restriction mimetics and cytochrome P450 metabolism are also areas of interest.
